"Another weekend without makeup" I think refers to how since there's no reason to get all done up, because she isn't going anywhere or seeing anyone important. When she asks, "Where do you go when you finish work?" she's accusing the other person, and "I don't like giving you the third degree" makes me think that she feels bad about suspecting, but has to know anyway.
